9 Analysis and Synthesis of Psychedelic Phenethylamines

Psychedelic phenethylamines are often used for the illegal drugs. We tried to analyze six psychedelic phenethylamines simultaneously. However, the standards were not easily obtainable. Therefore, they were synthesized and investigated by thin-layer chromatography (TLC), ultraviolet (UV) spectrometry and high performance liquid chromatography (HPLC). The compounds were satisfactorily separated by HPLC, though TLC gave an unsatisfactory separation. Each compound could be identified by NMR and Mass spectra. These results suggested that it is possible to separate and identify phenethylamine analogs using the combination of these analyses.

10 Species Identification for Animal Origin Drugs by PCR-RFLP Method    - Placenta Drugs -

The establishment of a method for species identification in medicinal products of animal origin has been desired, because usage of cattle-origin materials has been controlled since the bovine spongiform encephalopathy (BSE) problem occurred. We report here a simple method of molecular biological technique. DNA isolated from each animal specimens were used to polymerase chain reaction (PCR) and an approximately 360 bp fragments of the mitochondrial cytochrome b gene region were amplified. The following restriction fragment length polymorphism (RFLP) method discriminated animal species; human, bovine, porcine and ovine. This method also allowed the detection of human origin in drugs containing placenta which were formed into tablets and granules.

14 Determination of Thyroid Hormone in Dietary Supplements by Solid Phase Extraction Method

A method using solid phase extract (SPE) cartridges was developed for the determination of thyroid hormone, 3,3',5-Triiodo-L-thyronine (T3) and DL-thyroxine (T4) in a dietary supplement. T3 and T4 were loaded on a Oasis HLB SPE cartridge. The cartridge was neutralized with 3 mL of 1 mol/L acetic acid and washed with 3 mL of 40% methanol. It was then eluted with 2 mL of methanol and T3, T4 were determined by liquid chromatography-mass spectrometry (HPLC/MS). The repeatability of this method was obtained with a CV value within 5%. After 24 h, amounts of T3 and T4 in the HPLC/MS solution obtained by the SPE method and the ethyl acetate extract method were 95% and 50%, respectively. The proposed method was simple, rapid, and allowed a satisfactory clean-up.

16 Identification of Aloe Species in Health Foods by TLC and HPLC Analysis

The juice and the integument of leaves of 3 Aloe species, Aloe vera, A. ferox, and A. africana, are not allowed for use as food according to the Pharmaceutical Affairs Law in Japan. On the other hand, whole leaves of A. arborescens can be used as food. The present study was designed to distinguish Aloe species by TLC and HPLC analysis. By comparison of the characteristic TLC spots or HPLC peaks, it was possible to distinguish the 4 species. Thus, the botanical species of Aloe in commercial food products can be identified by TLC and HPLC analysis.
